Someone actually asked the author for an explanation in the Twitter thread, and they wrote that it's deliberately vague and there's no correct answer.
However, to me it seems like the most straightforward interpretation would be that Yoshida's "loss" was about losing the game of shiritori, whereas Mako apparently concluded that the reason Yoshida approached her was to cheer her up, and thus Mako "lost" by succumbing to Yoshida's charm.
